On en trouve dans le g\u00e9nome de la plupart des eucaryotes (organismes dont les cellules ont un noyau), o\u00f9 elles jouent un r\u00f4le important dans le contr\u00f4le de l&rsquo;expression des g\u00e8nes. Ainsi, un microARN qui s&rsquo;associe \u00e0 sa cible, un ARN messager (not\u00e9 ARNm, cet ARN code une prot\u00e9ine), l&#8217;emp\u00eache d&rsquo;\u00eatre traduit en prot\u00e9ine.<\/p>\n<p>  <!--more-->  <\/p>\n<p>\u00a0<\/p>\n<\/p><\/div>\n<p>Cependant, on ignorait le d\u00e9tail de cette r\u00e9pression : s&rsquo;agit-il d&rsquo;un simple blocage, ou bien l&rsquo;association microARN et ARNm conduit-elle \u00e0 la d\u00e9gradation de l&rsquo;ARNm ? Deux \u00e9quipes, celle de Sergej Djuranovic, de l&rsquo;Universit\u00e9 John Hopkins, et celle de Ariel Bazzini, de l&rsquo;Universit\u00e9 Yale, toutes deux aux \u00c9tats-Unis, ont tranch\u00e9 le d\u00e9bat et ont pr\u00e9cis\u00e9 les \u00e9tapes de ce m\u00e9canisme essentiel par lequel des g\u00e8nes sont r\u00e9duits au silence.<\/p>\n<p>Le premier groupe s&rsquo;est int\u00e9ress\u00e9 \u00e0 la cin\u00e9tique des \u00e9v\u00e9nements dans des cultures de cellules issues de mouches drosophiles. Le second a \u00e9tudi\u00e9 le fonctionnement des microARN chez le poisson z\u00e8bre <em>(Danio rerio)<\/em>. Leurs travaux montrent que, d&rsquo;abord, un microARN bloque la traduction d&rsquo;un ARNm, puis, qu&rsquo;ensuite, cet ARNm subit une transformation nomm\u00e9e d\u00e9ad\u00e9nylation qui consiste en l&rsquo;\u00e9limination de la queue poly(A), une succession de nombreux nucl\u00e9otides de type ad\u00e9nosine (A) situ\u00e9e \u00e0 l&rsquo;une des extr\u00e9mit\u00e9s des ARNm. Cette queue poly(A) est essentielle au d\u00e9clenchement de la traduction (elle participe au recrutement du ribosome) et \u00e0 la stabilit\u00e9 de l&rsquo;ARNm : la dur\u00e9e de vie d&rsquo;un ARNm dans le cytoplasme est d&rsquo;autant plus longue que cette queue l&rsquo;est aussi. Aussi, son \u00e9limination conduite par le microARN entra\u00eene la rapide d\u00e9gradation de l&rsquo;ARNm.<\/p>\n<p>On en d\u00e9duit que le microARN agit gr\u00e2ce \u00e0 deux m\u00e9canismes ind\u00e9pendants, le blocage de la traduction et la d\u00e9ad\u00e9nylation. Cela afin de maximiser la r\u00e9pression de l&rsquo;ARN cible, et donc du g\u00e8ne correspondant.
